Objective To extract and test circulating microRNA(miRNA) in the plasma and serum. Methods We utilized TRI REAGENTR BD to extract the total RNA in the plasma and serum, and the RNA was determined by electrophoresis in 10g/L non-denatured agarose gel. The miRNA cDNA Synthesis Kit was used to reverse the cDNA molecular of miRNA, which was then tested by the real-time PCR method. Results We successfully extracted and tested the total RNA in the plasma and serum. The miRNA in plasma and serum was tested by the reverse transcription and real-time PCR method. Conclusion These results suggest that we have successfully extracted and tested the miRNA in the plasma and serum, which can be used for real-time PCR experiments. This has laid some experimental foundation for further clinical application and theoretical research of miRNA.
