ABSTRACT
Capecitabine loaded Eudragit S 100& L 100 Nanoparticles were investigated for the approach of colon-specific drug delivery for the treatment of cancer. Drug loaded eudragit nanoparticles were prepared by solvent displacement method. The preparation was optimized with various ratios of drug, surfactant, stirring time and stirring speed and characterized by particle size measurement, scanning electron microscopy, entrapment efficiency and differential scanning calorimetry. The particles were characterized for drug content, particle size and particle morphology by scanning electron microscopy(SEM) . Nanoparticles obtained were of spherical in shape with particle size ranging from 163 nm to 222 nm and high encapsulation efficiency (61-78%). The in-vitro releases of capecitabine from the nanoparticles were studied by simple diffusion cell apparatus in pH 7.4 and 6.8 for 24hours. The release behavior of drug from the polymer matrix followed a sustained release. The decrease in drug release compare to Eudragit S Formulation (ESF) to Eudragit L Formulation (ELF) was due to higher concentration of the polymer. The in-vivo studies were performed by oral administration of Capecitabine loaded Eudragit S100 & L 100 Nanoparticles and the maximum percentage of drug release was in colon when compared to intestine & stomach.
