Visualizing membrane turnover in trypanosomes

After incubation with colloidal gold, x-ray dense particles accumulated within (or on the surface of) the cells of the trypanosome Crithidia fasciculata. These particles appeared to accumulate at the posterior of the cell and it is only when cells were imaged rapidly and after maintenance at 4°C that these particles were seen at the anterior of the cells. It would seem therefore that the internalization of the surface membrane is a very rapid process. Using an alternative photoresist material internal cellular organelles were successfully imaged.
