The article reports on the approval of the patent application for RecipharmCobra Biologics' novel plasmid maintenance system in Europe and the U.S. RecipharmCobra is given the privilege to own two plasmid maintenance systems including the Operator-Repressor Titration (ORT) through the invention of oriSELECT. According to vice president Simon Saxby, oriSELECT and ORT provide a state-of-the-art solution for antibiotic resistance gene transfer. Plasmids are replicating, circular DNA molecules that are naturally present in microorganisms.

Here we describe a facile plasmid-chromosome shuffling technique for generating and analyzing non-deletion alleles in the yeast Saccharomyces cerevisiae. This technique takes advantage of an existing set of genome-wide haploid-convertible heterozygous diploid yeast knockout mutants. This simple...

Background:CRISPR/Cas is a widespread adaptive immune system in prokaryotes. This system integrates short stretches of DNA derived from invading nucleic acids into genomic CRISPR loci, which function as memory of previously encountered invaders. In Escherichia coli, transcripts of these loci are...

While it has become increasingly clear that the Tree of Life hypothesis has limitations in its ability to describe the evolution of all evolving entities on the planet, there has been a marked reluctance to move away from the tree-based language. Ironically, while modifying the idea of the Tree...

The partition system of P1 plasmids is composed of two proteins, ParA and ParB, and a cisacting site parS. parS is wrapped around ParB and Escherichia coli IHF protein in a higher order nucleoprotein complex called the partition complex. ParA is an ATPase that autoregulates the expression of...

The conditions under which plasmids are predicted to persist remain controversial. Here, we reevaluate the ordinary differential equations used previously to model plasmid persistence and conclude that the parameter space required for maintenance is far less stringent than has been supposed....

Pilar; Toleman, Mark; Walsh, Timothy R.; de la Cruz, Fernando; Navarro, Ferran // Journal of Antimicrobial Chemotherapy (JAC);Jan2012, Vol. 67 Issue 1, p115

Objectives To gain insights into ampC transmission between bacterial strains. Methods We examined the genetic context of 117 acquired ampC genes from 27119 Enterobacteriaceae collected between 1999 and 2007. Plasmid analysis was carried out by PCR-based replicon or relaxase typing, S1-PFGE and...
