Flow injection-chemiluminescent assay for the determination of superoxide dismutase activity

ABSTRACT
Superoxide dismutase (SOD) activity was measured using a flow injection-chemiluminescent assay (FI-CLA) based on xanthine�xanthine oxidase dependent superoxide (O[sub 2] [sup .�] ) formation. The mobile phase consists of 50 mM potassium phosphate buffer (pH 7.4) containing lucigenin (5 �M) and xanthine (0.3 mM). Under optimum conditions, bovine albumin did not affect chemiluminescence at concentrations of 1�1000 �g mL[sup �1] and KCN inhibited 100 of the Cu, Zn-SOD activity using 5 �L of a 0.23 mM concentration. The analysis of one sample was done in less than 30 s with a relative standard deviation of �3.1. SOD activity in the biological samples was correlated to the amount of exogenously applied SOD. The FI-CLA method reported here appears to be one of the faster and more useful tools used to assay SOD activity.Key words: superoxide dismutase, flow injection-chemiluminescent assay, superoxide, xanthine oxidase, lucigenin.L'activit� de la dismutase du superoxyde (SOD) a �t� mesur�e par chimiluminescence � injection continue (FI-CLA) bas�e sur la formation de superoxyde (O[sub 2] [sup .�] ) par l'oxydase de xanthine. La phase mobile �tait constitu�e de 50 mM de phosphate de potassium (solution-tampon de pH 7.4), de lucig�nine 5 �M et de xanthine 0.3 mM. Dans des conditions optimales, l'albumine de bovins n'affectait pas la chimiluminescence � des concentrations de 1 jusqu'� 1000 �g mL[sup �1] et le KCN inhibait l'activit� du SOD-Cu,Zn � 5 �L de solution 0.23 mM. L'analyse d'un �chantillon s'effectuait en moins de 30 s avec une d�viation standard de �3.1 . L'activit� du SOD dans les �chantillons biologiques a �t� corr�l� � la quantit� appliqu�e exog�niquement au SOD. La m�thode FI-CLA mention�e ici s'av�re l'une des m�thodes les plus rapides et un des outils les plus pr�cieux pour �valuer l'activit� du SODMots cl�s : dismutase du superoxyde, �valuation par chimiluminescence � injection continue, superoxyde, oxydase de xanthine, lucig�nine.[Traduit par la R�daction]
